> What the Lyo2040 roadmap says about pharmaceutical lyophilization: PAT adoption, controlled ice nucleation, digital twins and microwave freeze drying.

Pharmaceutical lyophilization, the freeze drying process behind many vaccines, biologics and antibiotics, is due for an overhaul. That is the argument made by Alina Alexeenko of Purdue University in her recent BioProcess Online column, [The 5 Shifts That Could Remake Lyophilization By 2040](https://www.bioprocessonline.com/doc/the-shifts-that-could-remake-lyophilization-by-0001), which sets out where pharmaceutical freeze drying stands today and what it needs to become by 2040.

The opening comparison gets to the point quickly. A typical mid level car carries more process analytical technology than a pharmaceutical freeze dryer costing ten times as much. If a hatchback can monitor its tyre pressure, the cabinet that dried your sterile injectable should be able to monitor its product temperature.

Alexeenko is the Reilly Professor of Aeronautics and Astronautics and Chemical Engineering at Purdue and founding co director of LyoHUB, the industry consortium behind the [Lyo2040 roadmap](https://engineering.purdue.edu/Frontiers/2024/manufacturing/lyohub2040-technology-roadmap-for-pharma-biotech-manufacturing), developed with input from more than 100 experts across pharma, equipment manufacturing and regulation.

## What is lyophilization and why it needs modernizing

Lyophilization removes water from a drug product by freezing it and then sublimating the ice under vacuum, leaving a dry cake that can be stored for years and reconstituted before use. It has been part of pharmaceutical manufacturing since blood plasma was dried for battlefield medicine in the 1940s, and it remains the default for products too fragile to survive liquid storage.

The problem, as Alexeenko frames it, is that the process itself has barely moved. Over 70% of the dried antibiotics on the WHO Essential Medicines list are supplied as lyophilized products, many running processes designed decades ago. Lyophilized products also represented 13% of the FDA drug shortage list in 2021. Modernizing freeze drying is a supply resilience issue, not just a plant efficiency one.

## The five shifts in pharmaceutical freeze drying

The Lyo2040 roadmap organizes the future of lyophilization into five areas, each with measurable targets.

### 1. Lyophilization for ATMPs and new modalities

Cell and gene therapies and lipid nanoparticle formulations make up roughly 20% of the global pipeline, yet most depend on frozen or ultra cold storage today. The roadmap targets room temperature stability for 20% to 60% of products by 2040 and fewer than 5% requiring deep cold.

### 2. Formulation strategies for freeze drying

Controlled ice nucleation, lyobeads and other formats beyond the conventional cake in a vial, and AI assisted formulation are the headline items. The AI part depends on the industry actually sharing standardized data sets, which today it largely does not.

### 3. Process analytical technology in lyophilization

This is the car comparison. Only about 2% to 5% of cycles run closed loop control today, against a target above 75% by 2040. A standout example from Sanofi researchers showed sublimation rates estimated from existing sensors alone, agreeing with TDLAS measurements to within about 5% from pilot through commercial scale. Sometimes the win is using the data you already have.

### 4. Sterility assurance and automated visual inspection

Isolator technology and robotic loading improve sterility but remove manually placed probes, pushing the industry toward non invasive monitoring. Automated visual inspection currently false rejects 10% to 25% of lyophilized vials, with a target below 2%. A good cake should not fail for looking slightly different.

### 5. Digital twins for lyophilization processes

Process twins that follow a cycle in real time and equipment twins that can flag failures before they happen, with credibility frameworks borrowed from medical device software and open source tools like LyoPRONTO building the shared foundation.

## Related lyophilization research worth reading

The Pharmaceutical Research commentary [Critical Needs and Opportunities for Advanced Manufacturing of Lyophilized Injectables](https://pmc.ncbi.nlm.nih.gov/articles/PMC12222488/), published in 2025 with co authors from the FDA and Pfizer, is the scholarly companion to the column. It separates incremental improvements, such as equipment characterization and controlled ice nucleation, from bigger changes including spin freeze drying, aseptic spray drying and microwave vacuum drying.

The most experimental branch is microwave assisted lyophilization. A 2025 Scientific Reports paper from the Purdue group working with Merck and IMA Life describes [randomized field microwave drying near 18 GHz with closed loop control](https://pubmed.ncbi.nlm.nih.gov/40148465/), tested on formulations including an attenuated live virus vaccine, with drying times cut substantially versus conventional freeze drying. The technology is being commercialized through [LyoWave](https://www.lyowave.com/news-press/purdue-researchers-bringing-new-freeze-drying-methods-to-market), a Purdue spinout in which Alexeenko holds equity and patents, a disclosure made openly in the literature.

Quality and standards work matters just as much. A 2026 [AAPS Open](https://link.springer.com/journal/41120/articles) paper by Evgeniia Vorozhbit, Steve Nail and Alexeenko on vial fogging found a solution creep threshold near 27 degrees Celsius, driven by Marangoni flow, showing that inspection failures often begin in formulation and fill design. Purdue and LyoHUB also drove [ASTM E3250](https://engineering.purdue.edu/Engr/AboutUs/News/Spotlights/2021/2021-1025lyophilization), the first consensus standard for freeze drying instrumentation.

## What it means for pharma manufacturing

The appeal of Lyo2040 is that it gives lyophilization measurable goals rather than vague aspirations. Some progress will come from new equipment and new drying technologies. Some may come from getting more out of the equipment already installed, and the Sanofi work suggests the second category deserves a closer look before the next capital request.
